Common mistakes in Biomolecules
- Calling sucrose a reducing sugar. Its glycosidic linkage ties up both anomeric carbons, so it does not reduce Tollens' or Fehling's reagent - unlike maltose and lactose, which do.
- Confusing the D/L designation with optical rotation. D and L describe configuration relative to glyceraldehyde; + and − describe the observed direction of rotation, and the two do not have to match.
- Mixing up the levels of protein structure. Primary is the amino acid sequence, secondary the alpha helix and beta sheet, tertiary the overall three-dimensional fold, and quaternary the assembly of multiple chains.
- Assuming denaturation breaks peptide bonds. Denaturation destroys secondary and tertiary structure while leaving the primary sequence intact - which is why it is sometimes reversible.
- Swapping the sugars in DNA and RNA. DNA contains deoxyribose and thymine; RNA contains ribose and uracil.

Why is sucrose not a reducing sugar when glucose and fructose are?
Because the glycosidic bond in sucrose forms between the anomeric carbons of both units, leaving no free aldehyde or ketone group to be oxidised. Maltose and lactose keep one anomeric carbon free, so they still test positive.
